The 3M Petrifilm Rapid S. aureus Count Plate is a sample-ready culture medium system which consists of a Petrifilm Rapid S. aureus Plate and a Petrifilm Thermostable Nuclease Reactive Disk (TNase reactive disk). The Petrifilm Rapid S. aureus Plate contains modified Baird-Parker nutrients with a cold-water-soluble gelling agent, and the required Petrifilm TNase reactive disk contains DNA, toluodine blue-O, and a tetrazolium indicator that facilitates colony enumeration and confirmation of the presence of a Staphylococcal thermostable nuclease.
Petrifilm Rapid S. aureus Count Plates and disks are useful for the enumeration of Staphylococcus aureus. They provide confirmed results in as few as 26 hours. The plate and disk is equivalent to the BAM three-plate Baird-Parker agar and single tube-coagulase method.
For 3M Petrifilm Rapid S. aureus Count Plates, detection of TNase is a confirmatory method for S. aureus. An indicator in the Petrifilm TNase reactive disk causes colonies to appear red or blue. A second indicator causes pink TNase zones to appear, which confirms S. aureus. After second incubation with the disk, count all pink zones regardless of size.
